Studies on Capsule Formation: III. Inhibition of Capsule Formation of Klebsiella pneumoniae (Friedländer's Bacterium) by an Agent Produced by a Soil Bacillus.

چکیده

The conditions under which Klebsiella pneumoniae forms its largest capsules were reported in an earlier publication (Hoogerheide, 1939). The underlying principle of that study was the hypothesis that capsular polysaccharide synthesis is brought about by the action of an enzyme system in the cell, the activity of which can be studied in the same way as that of other metabolic enzyme systems. In a second paper (Hoogerheide, 1940) the influence of electrolytes on this enzyme system was studied, and it was found that electrolytes, when added to the culture medium, bring about a marked inhibition in the production of capsular polysaccharide. While engaged in attempts to isolate from bacterial sources possible enzymes capable of hydrolyzing the capsular substance of Friedlander's bacterium (similar to the enzymes described by Dubos and Avery (1931) and by Sickles and Shaw (1934, 1935), which digest the polysaccharides of Pneumococcus types I, II, III and VIII), we isolated several strains of a bacillus which appeared to have the remarkable property of excreting a soluble agent. By means of this agent, the bacillus is able to prevent Friedlander's bacterium from forming capsules. The present paper describes the isolation of this soil bacillus and the preparation of extracts containing the active principle.

Immunological Relationships of Encapsulated and Capsule-free Strains of Encapsulatus Pneumoniæ (friedländer's Bacillus)

1. "S" strains of Friedländer's bacillus produce capsules, soluble specific substance, and are of exalted virulence. "S" strains are type-specific and react with only the type-specific antibodies of the homologous types. 2. Immunization with "S" cells induces the formation of antibodies which agglutinate type specifically, precipitate the corresponding soluble specific substance, and protect wh...

Two distinct sensing pathways allow recognition of Klebsiella pneumoniae by Dictyostelium amoebae

Recognition of bacteria by metazoans is mediated by receptors that recognize different types of microorganisms and elicit specific cellular responses. The soil amoebae Dictyostelium discoideum feeds upon a variable mixture of environmental bacteria, and it is expected to recognize and adapt to various food sources. To date, however, no bacteria-sensing mechanisms have been described. In this st...

Capsule and fimbria interaction in Klebsiella pneumoniae.

The capsular polysaccharide and type 1 fimbriae are two of the major surface-located virulence properties associated with the pathogenesis of Klebsiella pneumoniae. The capsule is an elaborate polysaccharide matrix that encases the entire cell surface and provides resistance against many host defense mechanisms. In contrast, type 1 fimbriae are thin adhesive thread-like surface organelles that ...

Signature-tagged mutagenesis of Klebsiella pneumoniae to identify genes that influence biofilm formation on extracellular matrix material.

Klebsiella pneumoniae causes urinary tract infections, respiratory tract infections, and septicemia in susceptible individuals. Strains of Klebsiella frequently produce extended-spectrum beta-lactamases, and infections with these strains can lead to relatively high mortality rates (approximately 15%). Other virulence factors include production of an antiphagocytic capsule and outer membrane lip...
